{"id":11696,"date":"2017-08-23T14:21:58","date_gmt":"2017-08-23T20:21:58","guid":{"rendered":"https:\/\/www.uchealth.org\/today\/?p=11696"},"modified":"2020-11-11T16:51:46","modified_gmt":"2020-11-11T23:51:46","slug":"alzheimers-study-finds-surprising-quick-gains-for-patients","status":"publish","type":"post","link":"https:\/\/www.uchealth.org\/today\/alzheimers-study-finds-surprising-quick-gains-for-patients\/","title":{"rendered":"Alzheimer\u2019s study finds surprising, quick gains for patients"},"content":{"rendered":"<div style=\"margin-top: 0px; margin-bottom: 0px;\" class=\"sharethis-inline-share-buttons\" ><\/div><p>A team of University of Colorado School of Medicine researchers has reported rapid improvement in cognition and self-care capabilities among patients who took a drug called leukine.<\/p>\n<p>While the ongoing, early-stage study is small with a focus on safety among patients with Alzheimer\u2019s disease, leukine has a long history: it\u2019s a growth factor protein the U.S. Food and Drug Administration approved in 1995, and has been used ever since to stimulate white blood cell production in bone marrow transplant patients.<\/p>\n<p>CU Rocky Mountain Alzheimer\u2019s Disease Center (<a href=\"http:\/\/www.ucdenver.edu\/academics\/colleges\/medicalschool\/departments\/neurology\/clinical\/rmadc\/Pages\/default.aspx\">RMADC<\/a>) scientists <a href=\"http:\/\/www.ucdenver.edu\/academics\/colleges\/medicalschool\/departments\/neurology\/Faculty\/Pages\/potter.aspx\">Huntington Potter, PhD<\/a>, and Tim Boyd, PhD, are leading the ongoing research, with help from RMADC Clinical Director <a href=\"http:\/\/www.ucdenver.edu\/academics\/colleges\/medicalschool\/departments\/neurology\/Faculty\/Pages\/woodcock.aspx\">Jonathan Woodcock, MD<\/a>, who leads the <a href=\"http:\/\/www.ucdenver.edu\/academics\/colleges\/medicalschool\/departments\/neurology\/clinical\/rmadc\/clinic\/Pages\/MemoryDisordersClinic.aspx\">Memory Disorders Clinic<\/a> at UCHealth University of Colorado Hospital. The team presented its results at the recent Alzheimer\u2019s Association International Conference in London.<\/p>\n<p>The two-year <a href=\"https:\/\/www.uchealth.org\/today\/promising-alzheimer-drug-study-set-to-kick-off-at-uch\/\">study<\/a>, funded by CU, private philanthropy and a grant from the Dana Foundation, launched in October 2016. It\u2019s a continuation of years of work focusing on the drug\u2019s safety and effectiveness with Alzheimer\u2019s patients. Of 32 patients enrolled through Woodcock\u2019s clinic and the University of South Florida, 13 received daily leukine injections for three weeks, with the rest receiving placebo injections of saline solution.<\/p>\n<p>When tested just 18 days in, the patients who got leukine scored better on a standard cognitive impairment <a href=\"https:\/\/en.wikipedia.org\/wiki\/Mini%E2%80%93Mental_State_Examination\">test<\/a>. They also fared better in surveys, filled out by caregivers, designed to establish a patient\u2019s ability to clean, groom, feed and otherwise care for himself. Those who got the placebo saw no notable changes on either front.<\/p>\n<p>\u201cWe did not expect to see an improvement in any cognitive measure in such a short period of time,\u201d said Potter, who leads the RMADC, in an email. \u201cThe finding is preliminary and must be confirmed in a new trial, which we have received partial funding for from the Alzheimer\u2019s Association. The Alzheimer\u2019s Association estimates that 5.5 million people have the disease in the United States alone, about the same as the entire population of Colorado. The association <a href=\"https:\/\/www.cnn.com\/2017\/03\/07\/health\/alzheimers-report-2017\/index.html\">estimates<\/a> the U.S. cost of care at $259 billion a year, and that doesn\u2019t count the estimated $230 billion in unpaid care by family and others. With baby boomers heading into old age, the costs \u2013 financial and otherwise \u2013 are, without a medical breakthrough, poised to skyrocket.<\/p>\n<p>If leukine turns out to be that breakthrough, Potter and Boyd will be primarily to thank. They and colleagues published work that showed mice with Alzheimer\u2019s disease cleared their brains of 55 percent of disease-associated amyloid beta plaques after receiving leukine, and that the mice\u2019s cognition returned to normal.<\/p>\n<p>Leukine\u2019s exact effects on the brain remain unclear, but the researchers suspect a couple of benefits. One is that, in mice, it\u2019s a growth factor for neural stem cells. It also helps regrow blood vessels, which may be decisive, Potter and Boyd say: drugs that just clear out plaques leave voids that can induce brain swelling and microhemorrhaging. It may be that leukine\u2019s growth-stimulation helps the brain fill the voids with new blood vessels and\/or neurons, avoiding such damage.<\/p>\n<p>The team\u2019s prior leukine research extended to humans, too. They\u00a0<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/22905341\">studied<\/a>\u00a0the cognition of bone marrow transplant patients who had taken leukine for the standard three-week course of injections. They found that, six months later, such patients showed improved cognition and function as compared to those treated only with a different immune-system booster.<\/p>\n<p>In 2014, Potter and Woodcock\u00a0began <a href=\"https:\/\/uchealth-wp-uploads.s3.amazonaws.com\/wp-content\/uploads\/sites\/6\/2017\/02\/28144649\/alzheimers20leukine20trial.pdf\">assessing<\/a> the safety of leukine in Alzheimer\u2019s patients who took the drug for three weeks \u2013 the standard course for bone marrow transplant patients. The Alzheimer\u2019s patients have shown no adverse effects.<\/p>\n<p>It\u2019s been a long road to this point, and there\u2019s still a long road ahead. The history of Alzheimer\u2019s disease drug research is one with a prevailing theme is one of dashed hopes.
